To make oogonia, researchers differentiate human induced pluripotent stem cells (hiPSCs) into incipient mesoderm-like cells (iMeLCs) as adherent culture, then plate the iMeLCs in three-dimensional aggregates to generate human primordial germ cell-like cells (hPGCLCs). To generate xenogenic reconstituted ovaries (xrOvaries), researchers sort hPGCLCs from the aggregates and combine them with female mouse gonadal cells isolated from mouse embryonic (E) ovaries at E12.5. The xrOvaries are maintained in culture for 120 days, leading to hPGCLC-derived oogonia. The steps required for in vitro gametogenesis (IVG), including transition from hPGCLC-derived oogonia to oocyte and ultimately fertilization-competence, remain to be determined.
